Duties: We are seeking a Research Specialist B/C to work in the Cardiovascular Institute at the University of Pennsylvania Perelman School of Medicine. The role of this position is lab manager through coordination and support of the basic needs of the research group including but not limited to ordering lab supplies, interacting with vendors and service providers to maintain lab equipment and ensuring lab compliance to all NIH and PENN regulations.

Position duties for Research Specialist B:
1) Molecular Biology benchwork as assigned: Nucleic Acid and Protein Isolation/Purification, Western blotting, Southern blotting 25%
2) Maintain a research lab mouse colony through record keeping, animal husbandry, breeding, ear tagging and genotyping 25%
3) Daily maintenance tasks for the lab: Make stock solutions, ordering, inventory/package receiving, University Compliance procedures/updating protocols for IACUC, EHRS, Radiation Safety 25%
4) Perform research technical assistance to projects in the laboratory as assigned by supervisor 20%
5) Other duties as assigned 5%

Position duties for Research Specialist C:
1) Conduct independent research projects, analyze data and prepare manuscripts for publication 45%
2) Nucleic Acid and Protein Isolation/Purification techniques including bacterial culture preparation,DNA/RNA extraction, Quantitative Real Time PCR, Western blot analysis, Gel Electrophoresis, Northern blot analysis, Coomassie stains, Silver Stains, Affinity chromatography isolation of GST purified proteins 20%
3) Plasmid and transgenic mouse targeting construct design and validation including designing specific nucleic acid primers, restriction enzyme digest analysis, DNA sequencing analysis, vector backbone preparation /DNA ligation and validation of construct using Southern blot and PCR analysis 5%
4) Perform sterile cell culture techniques on primary cells, embryonic stem cells, in vivo lung explants and established cells culture lines including cell passaging, freezing down cells and cell differentiation assays, 3-D organotypic culture assays, DNA transfection/creation of stable cell lines expressing exogenous DNA, Soft Agar colony assays, Transmigration assays, Dual Gene Reporter assays 5%
5) Perform animal experiments including embryonic and adult microdissections of heart and lung tissue, immunohistochemistry, whole mount analysis of embryos, Lac Z staining, H&E staining, Immunofluorescence and histological tissue and slide preparation 5%
6) Supervise graduate students and additional personnel in teaching lab protocols and procedures 5%
7) Maintain a common lab mouse colony through record keeping, animal husbandry, breeding, ear tagging and genotyping through PCR and gel electrophoresis and complete the paperwork and parasitology testing necessary for animal transfers, imports and exports to/from our colony 5%
8) Prepare and maintain laboratory stock reagents, solutions, and common supplies and maintain lab inventory and equipment with preventative maintenance and cleaning, initiating service calls for repairs, training new users, maintaining lab manuals 5%
9) Other duties as assigned 5%

Qualifications: Research Specialist B- Bachelor's degree with a major in a science field that can be chemistry, biology, molecular biology, biochemistry, physics or related field and 1-3 years of hands-on laboratory experience or an equivalent combination of education and experience is required. The successful candidate must have a solid background in mammalian cell culture and molecular and cell biology techniques. Excellent verbal and written communication skills and understanding of scientific literature are required. Familiarity with computer programs including MS Office, Adobe Creative Suite is required. Strong organizational skills are necessary. The successful candidate will thrive in a team environment and is eager to learn new methodologies.

Research Specialist C-Minimum of a Bachelor's degree with a major in a science field that can be chemistry, biology, molecular biology, biochemistry, physics or related field and 3-5 years of hands-on laboratory experience or an equivalent combination of education and experience is required. Master's degree is preferred. The successful candidate must possess extensive experience in cell culture, molecular and cell biology techniques and previous experience with Fluorescence Activated Cell Sorting (FACS), qRT-PCR, immunocytochemistry. Excellent verbal and written communication skills and understand of scientific literature are required. Familiarity with computer programs including MS Office, Adobe Creative Suite is required. Strong organizational skills and ability to work independently is necessary. Will be responsible for conducting independent research projects and coordinating projects of other research personnel in the lab. The successful candidate will thrive in a team environment and is eager to learn new methodologies.
