The Division of Rheumatology has an opening for a full-time Post Doctoral Fellow position.

Overall Description of Responsibility or Nature of Work

This position reports directly to Dr. Kristen Demoruelle in the Division of Rheumatology. Dr. Demoruelle's research program investigates the immunobiology of the lung in association with pulmonary fibrosis in patients with rheumatoid arthritis (RA) with a particular focus on neutrophil and macrophage dysregulation. This position will be responsible for independently designing and conducting experiments that will establish the effect of specific proteins of interest on various lung cells obtained from human subjects with RA-associated interstitial lung disease (RA-ILD). The project will focus on the effects of citrullinated proteins, bacterial proteins and cytokine signaling pathways that have been previously identified to be of interest based on Dr. Demoruelle's prior studies. Cells of interest include neutrophils, macrophages and fibroblasts from the lung. The project will primarily use induced sputum samples from human subjects and human-derived cell lines along with cell culture, cell staining, and ELISA techniques. The project will compare changes induced in different lung cells between RA patients with and without ILD.

Specific Examples of Work

General daily activities:
    Plan and conduct day-to-day experimental activities related to the research project Collect, prepare and analyze research data including summarizing findings for publication of results Search pertinent scientific literature as needed Ensure quality control of all experiments and reagents Retain accurate records of research findings, along with analysis of results, in a detailed up-to-date electronic laboratory notebook Follow safe laboratory practices including BSL2+ level procedures for processing of human sputum samples Maintain clean and organized lab space Communicate effectively and collaboratively with other members of Dr. Demoruelle's research team

Ex vivo experiments:
    Isolate neutrophils and macrophages from induced sputum samples from human subjects using flow cytometry, microfluidics and other techniques Initiate and maintain primary and secondary cell cultures Stimulate cells in culture with multiple stimuli and inhibitors, including appropriate positive and negative controls, and quantify cell production of multiple cytokines and proteins Perform Western blot to measure activation of intracellular signaling pathways Perform immunostaining and RNA sequencing to determine changes in cell phenotype and function Establish protocols and optimal conditions for cryopreservation of cells

In vitro experiments:
    Perform ELISA and multiplex assays to measure cytokine and antibody levels Develop new ELISA assays to measure antibody reactivities to proteins of interest

General lab skills:
    Sputum and blood processing ELISA and multiplex assays Cell culture Western blot RT-PCR Immunofluorescence microscopy Flow cytometry

Special conditions of employment:
    Irregular hours may be necessary for some experiments.

Salary and Benefits

The salary range (or hiring range) has been established at $54,840- $66,600

Qualifications
Minimum Requirements

Applicants must meet minimum qualifications at the time of hire.

Graduation from an accredited college or university with an MD or PhD

Minimum of 3 years' experience working in a wet laboratory with basic laboratory skills

Experience with cell culture, cell staining and Western blot techniques

Desired or preferred

Experience with flow cytometry, RNA sequencing, immunofluorescence microscopy and ELISA assay development

Experience with Microsoft Word, Excel, Powerpoint, and Graphpad Prism

Knowledge, skills, and abilities

Excellent organizational skills

The ability to work independently within the research lab
