Duties/Responsibilities:

Molecular Biology & Microbial Growth: Design and construct expression plasmids of wild type and mutant forms of electron bifurcating Bfu enzymes in thermophilic and anaerobic microbes, grow them in large scale cultures (up to 600 L), and harvest the biomass.

Protein Chromatography: Starting with the microbial biomass and using activity-based and metal-based assay, purify the oxygen-sensitive metal-containing Bfu enzymes using multistep column chromatography.

Protein Assays: Characterize the purified Bfu enzymes proteins by carrying out biochemical and kinetic analyses.

Bioinformatics: Perform bioinformatic analyses of genome sequences of DNA from both metagenomes and pure organisms to assess Bfu enzyme distribution and phylogenetic relationships to inform growth and physiology studies. Approaches involve KEGG and BLAST analyses and basic metabolic modeling.

Position Summary:

To carry out microbial, genetic, and biochemical-based research as part of a team-based project using microbiology, biochemical, and bioinformatic approaches. The individual will characterize new types of enzymes that will be provided to internal collaborators within their own laboratory and external collaborators for additional structural-based analyses.

Relevant/Preferred Education, Experience, Licensure, Certification in Position:

Ph.D. in Microbiology, Genetics, or Biochemistry with a strong background in biochemistry and molecular biology.

Knowledge, Skills, Abilities and/or Competencies :

A Ph.D. in Biochemistry or related-field is needed. Have 1+ years experience in the characterization of anaerobic microorganisms and in protein purification. Have 1+ years experience in the genetic and bioinformatic analyses. Have experience in drafting, editing, and preparing scientific manuscripts and grants.

Physical Demands:

Working long hours on his/her feet at a laboratory bench. Occasional work on night and weekends maybe needed. Push heavy carts of glassware or gas cylinders. Be comfortable working with hazardous gases and chemicals regularly; and be comfortable wearing the required personal protective equipment. Duties involve attention to detail and are occasionally repetitive.
