We value the ability to engage effectively with students, faculty and staff of diverse backgrounds.Overall Description of Responsibility or Nature of Work:
This position reports directly to Diego Restrepo, Professor, Cell and Developmental Biology, and is responsible for studying the expression of olfactory receptor transcripts in the human olfactory bulb. This work is funded under an NIH BCCIN grant. It will use iterative amplified RNA-FISH labeling by fluidic exchange, we will initially image 130 RNAs and detail plans to expand the number of RNAs. The tissue will be imaged with high-speed single objective light-sheet microscopy for 3D imaging of proteins and RNA in the human olfactory bulb. we will develop a Bayesian nonparametric image analysis framework that self-consistently and simultaneously determines the probability associated with all RNA locations, numbers, and identities in the presence of variable autofluorescence and variable readout efficiency. Within the Bayesian paradigm, we propose a new approach to error correction in barcoded fluorescence experiments that significantly reduces the number of rounds required. The aim is to generate a 3D atlas of the transcriptome of the human olfactory bulb. Specific Position Duties with Percentages of Time:
[ 75 ]% Research and troubleshoot the iterative amplified RNA-FISH labeling technique and perform complementary immunohistochemistry.[ 25]% Assist graduate students and other post-doctoral fellows and technicians in the lab as instructed.Participate in the writing and editing of scientific manuscripts and grant applications.
