Position Summary:The McCown laboratory in the UNC Gene Therapy Center seeks to recruit a Senior Research Associate in order to advance the laboratory’s focus upon adeno-associated virus (AAV) vector properties that determine cellular transduction and gene expression in the rat central nervous system. This NIH funded, senior research position requires the ability to devise, conduct and evaluate experimental protocols that investigate the role of AAV vector elements which determine cell specific in vitro and in vivo transduction and gene expression.
Educational Requirements:Candidates must have a PhD or MD in the field of genomics or virology and a minimum of 3 years post-doctoral experience with a strong first author publication record involving AAV vector manipulation for in vivo CNS transduction and gene expression
Qualifications and Experience:Applicants are required to have extensive experience with structural manipulation of diverse AAV capsids, ability to identify specific functional elements of the AAV capsid, suspension and adherent cell production of recombinant AAV virus and analysis of outcomes of the specific capsid mutations. The applicant also should exhibit a knowledge of key elements that modulate in vitro and in vivo transcription activity, such as histones, transcription elements and miRNAs. With regard to specific techniques, the applicant should have an exhibited proficiency with AAV titer qPCR, AAV libraries, gel electrophoresis, western blotting, cell culture, brain tissue sectioning, immunohistochemistry, confocal microscopy. Given the need for an established track record in manuscript preparation, editing and publication, preference will be given to those applicants that have first author published manuscripts that address AAV capsid properties.
