Read CU Anschutz Quick Facts here.Nature of Work:A full-time (1.0 FTE) Professional Research Assistant (PRA) position is available in the area of pediatric heart failure research. The PRA will have a key role in conducting a variety of experiments exploring the molecular changes present in children with cardiomyopathy and heat failure. Laboratory techniques include, but are not limited to: RNA and protein extraction, cDNA synthesis, RT-qPCR, Western blotting, ELISA assays, maintaining primary cell culture, and other cell culture systems, mitochondrial functional assays, respirometry and may also include radioactivity assays for specific enzyme studies. Various models of heart failure have been developed, including primary cardiomyocyte cell culture as well as rodent models. Experiments are also performed utilizing human samples (Institutional Review Board approved Pediatric Cardiac Tissue and Blood Bank).Examples of Work Performed:- Rat ventricular myocyte preparation and culture
- Other specialized cardiomyocyte cultures- H9C2 cells
- Transfection and siRNA treatment of cells
- RNA, DNA, and protein extraction from cardiac tissue and cardiomyocyte cell culture
- Peripheral blood mononuclear cell isolation and serum processing from human whole blood samples
- Cardiac protein quantification through Western blotting
- Blue-Native PAGE assays of mitochondrial proteins
- RT-qPCR in a 384-well format
- Seahorse and Oroborus O2k respirometry
- Reagent preparation
- Organization and cataloging of human samples (in RedCap), antibodies, and chemicals
- Laboratory supply ordering and invoice tracking
QualificationsMinimum Qualifications:
- A baccalaureate degree in an appropriate scientific field (biology, molecular biology, biochemistry, physiology, pharmacology, genetics or similar
- Two years of full-time molecular biology laboratory bench research experience
- Hands-on experience handling pediatric cardiac tissue and processing pediatric blood samples
- Must successfully pass a drug test through Children’s Hospital Colorado
- Must be willing and able to pass a national criminal background check
- Training in molecular techniques with a good theoretical understanding and experience in interpretation of results
- Training in protein analysis techniques
- Cell culture techniques, including maintaining primary cell cultures
- Laboratory management skills
- Excellent organizational and documentation skills
- Detail-oriented and responsive
- Ability to collaborate and work effectively in a team environment
- Self-motivated and scientifically inquisitive
- Problem-solver
- Ability to communicate effectively, both in writing and orally
- Ability to work in the laboratory on weekends, as dictated by experiment schedule
