The Department of Pediatrics, Section of Genetics & Metabolism has an opening for a full-time (unclassified) Professional Research Assistant (PRA) position.

Nature of Work:

Research studies in mitochondrial functional assays. The person will grow fibroblasts and evaluate them with testing of mitochondrial biology such as enzyme activities of respiratory chain complexes, ATP synthesis, western blotting, and high resolution respirometry, and microscopy of in situ mitochondrial functional assays. These tests will be applied to normal and to mitochondrial disease specific samples. The person may also be involved in studies proving the pathogenicity of new genetic causes of mitochondrial disease, using tests as outlined above. Verification of a genetic defect via sequencing may be needed, and transfection with a normal and mutant gene copy to prove pathogenicity may also be pursued. The person will maintain the collection of cell lines from patients with metabolic disorders, in particular mitochondrial disorders at the laboratory.

 

Examples of Work Performed:

  • Tissue culture of fibroblasts and other eukaryotic cells. Testing of cells for mycoplasma contamination. Developing primary cultures of fibroblasts or lymphoblasts
  • Perform enzyme assays of mitochondrial enzymes
  • Perform western blotting of various kinds (denaturing and non-denaturing)
  • Measure ATP synthesis using HPLC
  • Perform high resolution respirometry
  • Perform labeling of mitochondria with reagents such as mitotracker, mitochondrial membrane potential probes, or COX histochemistry and perform confocal microscopy and analysis
  • Create genetic vector and perform transfection experiments

    QualificationsMinimum Qualifications:

    • Bachelor of Science or higher in biology, genetics, biochemistry or a related field.
    • Experience working in a laboratory
    Applicants must meetminimum qualifications at the time of hire.

    Conditions of Employment:

    • Must be willing and able to successfully pass a 10-panel drug test
    • Must be willing and able to successfully pass a national criminal background check
    Preferred Qualifications

    • Experience with functional testing in mitochondrial biology such as high resolution respirometry or Western blotting.  
    • Experience with mitochondrial assays, such as enzyme assays, HPLC
    • Experience with microscopic exams
    • Experience with molecular analysis such as sequencing, vector generation, transfection of cells
    Competencies:

    • Tissue culture for eukaryotic cells in particular human fibroblasts
    • The ability to work independently within the lab on established mitochondrial functional assays
    • The ability to write lab methods and scientific papers.
    • Organizational skills to maintain a log of cell lines with results
    • Excellent communications skills using the English language
    • Able to work in a research group context including flexibility in scheduling hours given current access restrictions
