Departmental Overview
The Nayak lab studies the physiology and evolution of methanogenic archaea. The SRA will assist with performing routine molecular biology experiments towards the lab's goals of generating randomly barcoded transposon mutant libraries. Primary duties of the research technician will include performing standard molecular biology techniques such as PCR for DNA amplification, gel electrophoresis and Gibson assembly reactions. Other duties will include routine microbiology cultivation techniques and maintenance of lab equipment, reporting results and assisting in drafting manuscripts. Ideal candidates would have prior experience in working with microorganisms and performing transposon mutagenesis.

Responsibilities
Molecular biology - Routine experiments including but not limited to:
  • Designing primers to perform PCR amplification of DNA
  • Gel electrophoresis
  • Plasmid DNA extraction and DNA purification
  • DNA ligation using the Gibson assembly technology
  • Sample preparation for sequencing
  • Analysis of sequencing data.

    Cultivation of Microorganisms - Routine experiments including but not limited to:
  • Preparation of growth media and other reagents
  • Using the spectrophotometer to measure optical density and assay growth rates
  • Setting up liposome mediated transformation assays with Methanosarcina
  • Maintaining the anaerobic growth chambers

    Required Qualifications
  • B.S. in microbial biology and at least one year of experience working at the laboratory bench and/or equivalent experience/training.
  • The candidate should be familiar with routine lab instruments such as a thermocycler, measuring balance, spectrophotometer, and a microcentrifuge.
  • Familiarity with molecular biology techniques such as PCR, DNA extraction, plasmid purification and gel electrophoresis.
  • Familiarity with sterile techniques for the cultivation of microorganisms and follow procedures to prepare growth media, reagents and buffers.

    Preferred Qualifications
  • Prior experience cultivating microorganisms and performing transposon mutagenesis
    is strongly preferred.

    Salary & Benefits
    This position is a non-exempt, bi-weekly paid position. Hourly pay for this role is commensurate with experience within the range of $21.36 - $23.27, Steps 1.0 - 5.0.
