DESCRIPTION

Under general supervision, incumbent will be interacting and working with Transgenic/Gene-targeting Core Facility personnel to provide transgenic and knockout mice to investigators in a full-scale production setting. Responsibilities include: Microinjection of 0.5 day mouse embryos and blastocysts, implantation of injected embryos into recipients, independent mouse surgery and animal colony maintenance duties, tissue and embryo collection, isolation of fallopian tubes/uterus from hormonally primed donor female mice, embryo harvest from fallopian tubes and uterine horns, hormone injections in mice, procedures such as Cesarean-sections, vasectomies, euthanasia, and cryopreservation of mouse sperm.

Performing specialized embryonic stem cell tissue culture, generating knockout clones by transfection and selection of stem cells, generation of mouse embryonic fibroblasts from various antibiotic resistant strains of mice. Make important original contributions pertaining to improving experimental procedures. Evaluate ongoing experimental procedures on a continuous basis for efficiency and data significance, identify areas in need of changes and recommend modifications as needed. Additional duties include data management and lab support, and interaction with core clients, Principal Investigators, Core Manager Animal Care Program, and Core Directors.

MINIMUM QUALIFICATIONS
  • Advanced theoretical knowledge of molecular biology or related field and direct experience with genetic manipulation in mice; and/or comparable education and experience.

  • Demonstrated knowledge and experience in tissue culture and sterile technique.

  • Experience performing implantation surgery and other procedures with small animals such as C-section, vasectomy, and/or euthanasia.

  • Experience working with and handling mice. Extensive experience with priming of mice and embryo collection and evaluation.

  • Demonstrated experience organizing and maintaining mouse colonies.

  • Advance knowledge of mouse reproduction and embryology.

  • Working knowledge of nucleic acid chemistry, recombinant DNA manipulations and gene transfer.

  • Demonstrated experience analyzing and interpreting data.

  • Strong working knowledge of various software applications such as Microsoft Word, Excel, Filemaker Pro, and Adobe Photoshop to be able to contribute to completing grants, manuscripts, and presentations.

  • Excellent communication skills (verbal and written) needed to interact professionally and effectively in work environment, to effectively and accurately document research procedures. Ability to read, comprehend, and discuss research materials.

PREFERRED QUALIFICATIONS
  • Experience with micro-injection of DNA/RNA into the pronucleus or cytoplasm of 0.5 day embryo and injecting ES cells into blastocysts.
  • Demonstrated experience with Embryonic Stem cell (ES) culture and ES cell clone picking.
SPECIAL CONDITIONS

  • Must be willing and able to work with small animals including mice.
  • Must be willing to perform surgeries on live mice and sacrifice mice.
