Primary Function of Organization Unit: The USDA-APHIS-PPQ-S&T CPHST Beltsville Laboratory mission is to provide federal confirmatory diagnostics for regulatory plant pathogens; develop and validate advanced molecular and serological detection methods for pathogens that threaten U.S. agriculture; evaluate applicability of new, state-of-the-art technologies and methodologies in support of agency efforts.
Essential Job Duties: The incumbent will work on the project: Development and validation of advanced methods for detection and identification of plant quarantine pathogens. The incumbent will work under the general supervision of a Team/Project Leader to support the APHIS PPQ CPHST Beltsville Laboratory projects on High Throughput Sequencing (HTS) including 2nd Generation Sequencing (NGS) and 3rd Generation Sequencing (N-NGS). The incumbent will support the ongoing development, validation and troubleshooting of HTS assays, including targeted DNA-seq and Whole Genome Sequencing. The incumbent will provide support for the laboratory operations of different NGS workflows; from sample preparation, library construction, data generation, data processing to interpretation of results, ensuring high level of quality and reliability of generated results. The incumbent will draft experimental plans, validation reports, work instructions, technology transfer documents, and SOPs for the diagnosis of regulatory plant pests/pathogens. The incumbent will maintain detailed notebook laboratory records and train other staff as needed. One of the primary goals for the CPHST Beltsville Laboratory is the identification and characterization of plant pathogens of quarantine and regulatory significance. **This position is based at the CPHST Beltsville Laboratory in Beltsville, MD.**
Minimum Education/Experience: * Ph.D. or equivalent doctoral degree in Molecular Biology, or a related field. * Must have experience managing and working with Sanger and High Throughput Sequencing (HTS) including 2nd Generation Sequencing (NGS).
Department Required Skills: * Knowledge and experience of microorganisms culturing (fungi & Oomycetes), and DNA & RNA isolation techniques. * Experience with advanced lab equipment, cloning kits, library preparation for High Throughputs Sequencing (HTS) and Thermo cycler application. * Experience with NGS and N-NGS assay development, validation, troubleshooting, and reviewing of run QC data. * Knowledge of different HTS platforms and technologies [experience with Next-next Generation Sequence (long reads sequencing) is a plus]. * Technical expertise with other molecular technologies, including: PCR, quantitative real-time PCR, digital PCR, and Isothermal Amplification.
Preferred Experience, Skills, Training/Education: * Experience with 3rd generation DNA sequencing technologies. * Knowledge of technical operating systems, skilled in methods development. * Ability to plan and follow planned activities independently. * Excellent written and oral communication skills, strong work ethic, emphasizing both efficiency and quality of work. * Training in molecular and microbiological diagnostic techniques using HTS analysis will be an added advantage. * The incumbent should have ability to utilize their knowledge of microorganisms, genetics, and genomics in diagnostic applications.
