Nature of Work:
A Professional Research Assistant (PRA) full-time position is available in the laboratory of Dr. Melanie Knigshoff in the Department of Pulmonary Sciences and Critical Care, University of Colorado School of Medicine. The lab investigates the role of developmental pathways, such as Wnt signaling in chronic lung disease, specifically how developmental signal mediators are involved in lung injury, disturbed epithelial-mesenchymal crosstalk, and extracellular matrix deposition in idiopathic pulmonary fibrosis (IPF) and chronic obstructive pulmonary disease (COPD).
Duties and Responsibilities:
The successful candidate will be a productive member of the research team, participate in research projects, and help maintain the operation of the laboratory. They will also be expected to develop and maintain computational research, conduct literature searches to design experiments and troubleshoot protocols, record and analyze research data, and assist with preparing research manuscripts and presentations. Experience and background in molecular biology, skilled in tissue culture work, PCR, qPCR and western blot applications are mandatory. The applicant needs to be able to work independently and contribute as a self-motivated member of the team. The employee will be working closely with the principle investigator.
Practical experience and advanced knowledge in one or more of the following techniques is preferred:
- Advanced microscopy techniques; confocal, time-lapse imaging
- Histology - immunofluorescence staining, paraffin embedding
- Primary cell isolations and culture (mouse and human tissue), organoid culture
- Classical molecular biology readouts: PCR, qPCR and Western blotting applications
- Luciferase assays, protein and RNA extractions
- Flow cytometry and FACS analysis
Other responsibilities within the lab may include:
- Perform experiments and tabulate/analyze research data
- Provide animal care and technical support for mouse related research procedures
- Review, develop and implement new or modified laboratory techniques required for research projects with potential to lead independent research project
- Training of laboratory personnel (graduate students and visiting scientists) in experimental techniques
- Present data during laboratory meetings and participate in journal club discussions

This position requires a Bachelor's Degree in Biology, Chemistry, Microbiology or a similar field and 3-5 years of work experience with associations to publications. The ideal candidate must have experience with one or more of the following skills: advanced microscopy techniques, primary cell culture, or flow cytometry and FACS analysis, as well as molecular and cell biology readouts.
