Job Summary
Performs a variety of technical duties involved in conducting laboratory tests, experiments, and determinations in support of Duke University research operations.

Work Performed
  1. Performs a variety of complex technical duties involved in conducting physical, chemical, biological, and other research laboratory tests, experiments, and determinations to obtain data for research purposes; compiles, analyzes, documents, and draws tentative conclusions from experimental results. Confers with principal investigator to review work assignments and develop plans for research experiments; makes minor modifications to existing laboratory procedures and techniques to meet the needs of the particular equipment as required. Reviews the laboratory methods manual, scientific journals, abstracts and other literature for information applicable to research experiments.
  2. Researches mouse strains genotypes and phenotypes to develop the best practice specific to strain in regards to breeding schemes. Develops plan to deliver animal subjects to Core investigators for experimental use tailored to investigator needs. Develops timelines in regards to colony production for Core investigators. Tracks pedigrees of strains. Develops strategy for maintenance of genetic purity in lines. Communicates with Core investigators regarding their lines and study needs. Communicates with Breeding Core manager regarding censuses, line specific concerns, and maintenance of lines. Researches white papers in regards to current techniques in breeding, cryopreservation, PCR, genotyping, phenotyping, superovulation, and collection of gametes from donor mice.
  3. Determines and sets up instruments, materials, and apparatuses; operates laboratory equipment required for specific tests.
  4. Sets up instruments for PCR (thermal cycler, electrophoresis, gel camera) and associated media such as microscopy, sperm analysis (visual and computerized), operation and maintenance of liquid nitrogen freezers, anesthesia machines, instrument cleaning and sterilization tattoo machine.
  5. Obtains, prepares, mounts, and stains tissue, blood, and other substances for microscopic examination.
  6. Performs tail biopsies, ear punch biopsies, blood collection for genotyping, as well as organ and fecal collections for investigators.
  7. Prepares culture media, chemicals, reagents, and solutions related to laboratory procedures.
  8. Maintains sufficient inventory of materials, supplies, and equipment for performance of duties; cleans and maintains laboratory equipment.
  9. Maintains inventory of animals.
  10. Reviews work of lower level laboratory personnel within designated areas and assists in training new personnel.
  11. Performs on-call requirements as assigned which include weekend and holiday rotations.
  12. Performs other related duties incidental to the work described herein.


Skills
  1. Clinical observations & evaluations of animal health status
  2. Assist in surgery
  3. Anesthetize, catheterize, and intubate a variety of animals
  4. Proficient at basic rodent technical skills (IP, SQ, IV injections, blood collection, etc.) 5. Administer daily treatments and medications
  5. Basic animal restraint techniques
  6. Basic diagnostic lab techniques
  7. Basic radiograph techniques
  8. 9. Participate in enrichment plan
