Occupational Summary

Conduct a variety of highly technical experimental procedures and other tasks, including imaging with the latest confocal microscopes, involved in the planning, development and implementation of a research project at Duke University. Work Performed Perform experimental procedures involving multi-step or complex laboratory methods to collect data and new information for a Duke University research project, by operating specialized, highly technical laboratory instruments and equipment, where necessary. Devise or modify experimental techniques for tests and analyses. Maintain detailed laboratory notes, documenting procedures, results and interpretations of findings, in compliance with the Duke University laboratory SOP and Duke University requirements. Evaluate and interpret data, and prepare reports and analyses of progress, adverse trends, recommendations and conclusions. Prepare specimens for live and fixed cell imaging, and collect images using light and fluorescence microscopes available in the Duke University laboratory and the Duke University LMCF, including recent/latest model confocal and other microscopes. Stain fixed tissue with antibodies and localize antibodies by imaging. Analyze live and fixed images using image analysis software, including ImageJ/FIJI, to obtain data for publication. As needed, obtain TEM images in the Duke University Electron Microscopy Facility, analyze images and prepare images for publication. Use molecular biology techniques to perform experiments needed to carry out the research project, including DNA/RNA extraction, plasmid preps, PCR, gel electrophoresis, molecular cloning, transformation, Southern blots, protein preps and Western blots. Perform cell culture, transfection, live imaging or fixation of cultured cells, and antibody staining of cells, as necessary. Contribute to the development of procedures involving CRISPR/Cas9 mutagenesis and gene modification. Assist with Drosophila culture as needed, including stock maintenance and genetic crosses. Review journals, abstracts and scientific literature to keep abreast of new developments and to obtain information regarding previous experiments to aid in the design and development of new procedures and techniques. Confer with principal investigator in determining specific experiments to be included in the research protocol and to discuss the interpretation of results. Supervise, train and instruct students or other personnel in laboratory techniques and procedures. Perform laboratory management duties, including ordering of supplies and maintaining inventories, laboratory safety and hazardous substance requirements, and recombinant DNA compliance.

Education
