Research Analyst I position at Duke Viral Vector Core
The Duke Viral Vector Core is a state-of-the art facility that provides comprehensive set of viral services to the Department of Neurobiology and Duke research community. We are currently looking for a Research Analyst I, who will help us to support ongoing service and research programs. Essential Duties and responsibilities. Design and exercise various strategies for engineering and cloning new viral constructs, aimed to enhance packaging and expressional characteristics of adeno-associated and lentiviral-based vector systems. Generate/expand a collection of DREADDs and other optogenetic-expressing constructs
And vectors. Lead project aimed to develop novel gene-editing viral vector tools for high-throughput knockout of
human genes. Carry out molecular cloning and DNA manipulation, and additional molecular biology tasks to support ongoing service and research programs of the core. Assume responsibility for production, QC/sequencing, tittering, and tracking of plasmids and vectors. This will involve preparation of DNA for large-scale vector production using commercially available kits, evaluation of their concentration and purity and routine tissue culture work. Assist in general service operations of the Duke VVC. Communicate effectively with the PI and other members of the team. The person will establish work flow with the help of principle investigator and expected to execute procedures and experiments independently with minimal supervision. The person will be expected to supervise and help the work of lower level employees, such as students and train new personnel.

Preferences, Education/Experience
PhD degree or equivalent; previous experience producing constructs for viral vector production (lentivirus, adeno-associated virus, HSV-1 or adenovirus) is a plus. Demonstrated, extensive experience with molecular biology techniques is required, including: PCR, restriction digestion & cloning, primer design, plasmid isolation and analysis, bacterial transformation and screening, extraction of genomic DNA, DNA sequencing and analysis. Accompanying data analysis software proficiency is required.

Education

Work requires a bachelor's degree in botany, biology, zoology or other directly related scientific field.
