The Clark Lab in The Institute for Stem Cell Biology and Regenerative Medicine seeks a skilled technician to help maintain transgenic mouse populations. This position is 100% FTE and will perform the following duties:
  • Maintain transgenic mouse populations, set up breeding and weaning; ear tag mouse and tail mouse for genotyping.
  • Digest mouse tail, extract DNA, set up and perform PCR, run agrose gel electrophoresis and interpret results for genotyping.
  • Maintain cell cultures, to prepare, stain and observe histology sections under microscope.
  • Learn and perform other more advanced tasks, depending on ability and time available, such as animal surgical procedures.
  • Maintain detailed record from genotyping and other experiments in words, charts and/or images, to analyze and interpret the results.

Required:
  • At least two years of previous laboratory experience or two years of college coursework in related scientific discipline required.
  • The incumbent will independently perform skilled techniques that require delicate manipulation and/or fine adjustments. S/He will draw upon previous laboratory experience with the test or equipment to detect if something is wrong with the procedure and may make procedural changes or modify techniques. S/he may be expected to complete general mathematical analysis. This position requires a high level of responsibility and independence. Work is performed routinely without direct supervision, however all work is reviewed and discussed with supervisor.
  • Solid written and verbal communication skills, as well as demonstrated professionalism in working with colleagues (both internal and external laboratory users).
  • Must be capable of and willing to pay strict attention to detail, including following written and oral instruction. Familiarity with the following experimental techniques desired:
    • a. Maintain and update transgenic mouse breeding scheme with detailed strain names, tag numbers, date of birth and date of weaning of parental or offspring mice on the cage card of each cage.
    • b. Maintain and update multiple transgenc mouse populations of various strain names and their genotypes from genotyping on computer spreadsheet.
    • c. Follow various established PCR reaction recipes of reagents and setup protocols of reaction conditions. May need occasional trouble-shooting or optimization with supervisor.
    • d. Making agrose gel and perform electrophoresis by following established protocol.
    • e. Maintain mammaian cell cultures by following established medium recipes, conditions and setup procedures, and by observing the images under microscope.
    • f. Perform fixing and staining of tissue sections by following established protocols and observing the quality under microscopy. May need to observe the images and make analysis if capable after training and practicing.
