Occupational Summary

Perform a variety of complex technical duties involved in conducting in vitro and molecular laboratory tests as well as molecular and animal model experiments in support of Duke University research operations.

Work Performed

Perform a variety of complex technical duties in a dynamic and well-funded laboratory dedicated to fungal pathogenesis and translational research (www.steinbachlab.org). This position will facilitate research for the lab faculty, post-doctoral fellows, and graduate students by conducting biochemical, biological, molecular, animal model (mice)and other research laboratory tests, experiments and determinations to obtain data for research purposes. This particular position has a large focus on the animal model studies, so experience in working with mice (handling, intraperitoneal and subcutaneous injections) is a requirement. There is no breeding involved.

Advanced participation in research projects will result in including this position on authorship in scientific publications. This position will compile, analyze, document and draw tentative conclusions from experimental results; confer with principal investigator to review work assignments and develop plans for research experiments; make minor modifications to existing laboratory procedures and techniques to meet the needs of the particular equipment as required; and review laboratory methods manual, scientific journals, abstracts and other literature for information applicable to research experiments.

Determine and set up instruments, material and apparatus and operate laboratory equipment required for specific tests. Perform mouse intraperitoneal and subcutaneous injections as well as intranasal and inhalational infection. Obtain mouse blood samples for testing, as well as organ harvesting via necropsy for analyses. Perform molecular studies related to Aspergillus fumigatus fungal pathogenesis, including in vitro antifungal susceptibility testing, DNA/RNA isolation, Southern blots, protoplasting transformation of DNA, PCR, and other molecular testing. Prepare culture media, chemicals, reagents and solutions for laboratory experiments. Maintain sufficient inventory of material, supplies and equipment (including ordering) for performance of duties; clean and maintain laboratory equipment. Review work of lower level laboratory personnel within designated areas and assist in training new personnel.
